Product Overview

Clone GIF-9 is a rabbit monoclonal antibody that recognizes insulin receptor substrate 1 (gene IRS1) in human samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 131.6 kDa, and the supplier reports an observed band at about 170 kDa.

Insulin receptor substrate 1 (IRS1) is an adaptor protein phosphorylated on tyrosine by the insulin and IGF-1 receptors. Phosphorylated IRS1 recruits PI3K and GRB2, relaying signals to the AKT and MAPK pathways that control glucose uptake, metabolism and growth. Inhibitory serine phosphorylation of IRS1 is a common readout in insulin resistance research.

Validated Applications

Western blot 1:1000–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200

Samples with a confirmed band (WB): HEK293 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Immunogen

Synthetic peptide based on the sequence of human IRS1.

Reactivity Notes

The supplier lists reactivity with human. UniProt places insulin receptor substrate 1 in the cytoplasm and nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:1000–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.Has this antibody been tested on canine samples?
A.Reactivity with canine samples has not been tested by the supplier. Aligning the immunogen sequence with the canine ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 131.6 kDa; observed band at about 170 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human IRS1.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
