Host Rabbit
Immunogen A synthesized peptide derived from human Islet1 ISL1 (ISL1 transcription factor, LIM/homeodomain) is a member of the LIM/homeodomain family of transcription factors. It binds to the enhancer region of the insulin gene, among others, and may play an important role in regulating insulin gene expression. It is central to the development of pancreatic cell lineages and may also be required for motor neuron generation. Islet-1 expression defines cardiac progenitor cell populations and is required for normal cardiac development and asymmetry.

Key elements and design rationale

  • Target: ISL1 (Insulin gene enhancer protein ISL-1).
  • Antibody format: Monoclonal; clone AFIA-9; isotype Rabbit IgG.
  • Host: Rabbit.
  • Species reactivity: Human (confirm in your model system with appropriate controls).

This description is intended to help interpret the antibody design and the biological context of the target using the fields provided in the catalog record, alongside general experimental considerations.

Biological background

ISL1 (protein: T-cell surface glycoprotein CD3 zeta chain) is a commonly studied target in molecular and cellular biology. Functional context (as provided): Binds and regulates the promoters of the insulin, glucagon and somatostatin genes. Involved in the specificarion of motor neurons in cooperation with LHX3 and LDB1 (By similarity). . Reported cellular localization context: Nucleus. Tissue expression notes (as provided): Expressed in subsets of neurons of the adrenal medulla and dorsal root ganglion, inner nuclear and ganglion cell layers in the retina, the pineal and some regions of the brain.

Research relevance and current trends

  • Research context keywords from the source record include: Neurogenesis,Neurology Process,Neuron Marker,Neuroscience,Organogenesis,Stem Cells,Transcription Factors/Regulators.
  • Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
  • Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.

Common research applications

  • Western blotting (WB): assess relative target abundance across samples, treatments, or time-points.
  • Immunohistochemistry (IHC): evaluate spatial distribution of target-positive staining in tissue architecture.
  • Immunofluorescence/ICC (IF/ICC): visualize subcellular localization patterns and cell-to-cell heterogeneity.
  • Flow cytometry: quantify target-positive populations and compare shifts in marker distributions.
  • Immunoprecipitation (IP): enrich target complexes for downstream immunoblot or interaction analyses.

Workflow ideas (metafield): Validate ISL1 antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect ISL1 expression by Western blot in cell or tissue lysates, Detect ISL1 in FFPE tissue sections by immunohistochemistry, Localize ISL1 by immunofluorescence/immunocytochemistry in cultured cells, Quantify ISL1-positive cells by flow cytometry in single-cell suspensions, Enrich ISL1 by immunoprecipitation from lysates for downstream analysis

Notes for experimental interpretation

  • Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
  • Apparent molecular weight may vary by sample type and processing (observed MW: 28 kDa; calculated MW: 39036 MW).
  • Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.

Additional product details (from the source record)

  • Molecular weight (observed): 28 kDa
  • Cellular localization (provided): Nucleus.
  • Tissue details (provided): Expressed in subsets of neurons of the adrenal medulla and dorsal root ganglion, inner nuclear and ganglion cell layers in the retina, the pineal and some regions of the brain.

I have a question about product M02969-1, anti-Islet1 Monoclonal antibody. I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?
It is not recommended storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free M02969-1 anti-Islet1 Monoclonal antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.
Will anti-Islet1 Monoclonal antibody M02969-1 work for IP with platelet?
According to the expression profile of platelet, ISL1 is highly expressed in platelet. So, it is likely that anti-Islet1 Monoclonal antibody M02969-1 will work for IP with platelet.
