Product Overview
This rabbit polyclonal antibody recognizes inositol-3-phosphate synthase 1 (gene ISYNA1) in human, mouse and rat samples and is validated for Western blot, IHC and ELISA. The predicted molecular weight is 61.1 kDa, and the supplier reports an observed band at about 61 kDa.
Inositol-3-phosphate synthase 1 (ISYNA1) catalyzes the NAD+-dependent conversion of glucose 6-phosphate to myo-inositol 1-phosphate. This is the rate-limiting first step of de novo inositol synthesis, which supplies inositol for phosphoinositide lipids and inositol phosphate signaling molecules.
| Target | Inositol-3-phosphate synthase 1 (gene ISYNA1; UniProt Q9NPH2, human) |

Validated Applications
| Western blot | 0.1–0.25 µg/mL (Human, Mouse, Rat) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human, Mouse, Rat) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human HepG2 cells, human K562 cells, human 293T cells, human HEL cells, rat testis tissue, mouse testis tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human glioblastoma tissue, human liver cancer tissue, human lung cancer tissue, human pancreas ductal adenocarcinoma tissue, human placenta tissue, human testicular seminoma tissue, human urothelial carcinoma tissue, mouse testis tissue, rat testis tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Recombinant human ISYNA1 fragment (Gln24–His454), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 91.6% and 92.8%, respectively.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places inositol-3-phosphate synthase 1 in the cytoplasm. Tissue expression noted by UniProt (human): Highly expressed in testis, ovary, heart, placenta and pancreas. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
