Overview

This antibody is intended for detection of JAK1 (Tyrosine-protein kinase JAK1) in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.

Vendor notes: Boster Bio Anti-JAK1 Antibody Picoband® catalog # A00330. Tested in WB applications. This antibody reacts with Human, Mouse, Rat. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: A synthetic peptide corresponding to a sequence at the N-terminus of human JAK1, different from the related mouse sequence by three amino acids.
  • Molecular weight context: reported MW: 133 kDa; calculated MW: 133277 MW
  • Reactivity: Human,Mouse,Rat
  • Applications: WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

Tyrosine-protein kinase JAK1; Tyrosine-protein kinase JAK1. JAK1 (JANUS KINASE 1) is a human tyrosine kinase protein essential for signaling for certain type I and type II cytokines. It is a member of a new class of PTKs that are a large family of proteins characterized by the presence of a second phosphotransferase-related domain immediately N-terminal to the PTK domain--hence the name Janus. The JAK1 gene is mapped to 1p31.3. JAK1 is also important for transducing a signal by type I (IFN-α/β) and type II (IFN-γ) interferons, and members of the IL-10 family via type II cytokine receptors. Additionally, Jak1 plays a critical role in initiating responses to multiple major cytokine receptor families. Loss of Jak1 is lethal in neonatal mice, possibly due to difficulties suckling. Expression of JAK1 in cancer cells enables individual cells to contract, potentially allowing them to escape their tumor and metastasize to other parts of the body. Functional note: Tyrosine kinase of the non-receptor type, involved in the IFN-alpha/beta/gamma signal pathway. Kinase partner for the interleukin (IL)-2 receptor. . Reported localization: Endomembrane system; Peripheral membrane protein. Wholly intracellular, possibly membrane associated. Expression/tissue context: Expressed at higher levels in primary colon tumors than in normal colon tissue. The expression level in metastatic colon tumors is comparable to the expression level in normal colon tissue. .

Research relevance and current trends

  • Apoptosis: Researchers commonly examine how JAK1 (Tyrosine-protein kinase JAK1) relates to this theme using model systems and orthogonal readouts.
  • Cancer: Researchers commonly examine how JAK1 (Tyrosine-protein kinase JAK1) relates to this theme using model systems and orthogonal readouts.
  • Epigenetics and Nuclear Signaling: Researchers commonly examine how JAK1 (Tyrosine-protein kinase JAK1) relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative JAK1 (Tyrosine-protein kinase JAK1) levels across conditions; band patterns may reflect isoforms and processing.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins.
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.

We have been able to see staining in mouse blood. Are there any suggestions? Is anti-JAK1 antibody supposed to stain blood positively?
From literature blood does express JAK1. From Uniprot.org, JAK1 is expressed in blood, brain, colon tumor, liver, among other tissues. Regarding which tissues have JAK1 expression, here are a few articles citing expression in various tissues: Brain, Pubmed ID: 15489334, 16710414 Colon tumor, Pubmed ID: 7896447 Liver, Pubmed ID: 24275569
I would like to test anti-JAK1 antibody A00330 on human liver for research purposes, then I may be interested in using anti-JAK1 antibody A00330 for diagnostic purposes as well. Is the antibody suitable for diagnostic purposes?
The products we sell, including anti-JAK1 antibody A00330, are only intended for research use. They would not be suitable for use in diagnostic work. If you have the means to develop a product into diagnostic use, and are interested in collaborating with us and develop our product into an IVD product, please contact us for more discussions.
