Product Overview
Clone 18J08 is a rabbit monoclonal antibody that recognizes tyrosine-protein kinase JAK1 in human, mouse and rat samples and is validated for Western blot. The predicted molecular weight is 133.3 kDa, and the supplier reports an observed band at about 133 kDa.
JAK1 (Janus kinase 1) is a cytoplasmic tyrosine kinase bound to many cytokine receptors, including those for interferons, IL-6 family cytokines and cytokines that use the common gamma chain. Upon cytokine binding, JAK1 phosphorylates the receptor and STAT proteins, which then regulate gene expression, and it is a major target of JAK inhibitor research.

Samples with a confirmed band (WB): human A431 cells, human A549 cells, rat RH35 cells, rat PC-12 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1: 500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places tyrosine-protein kinase JAK1 in the endomembrane system. Tissue expression noted by UniProt (human): Expressed at higher levels in primary colon tumors than in normal colon tissue. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
