Product Overview

This rabbit polyclonal antibody recognizes kallikrein-1 (gene KLK1) in human samples and is validated for ELISA and Western blot. The predicted molecular weight is 28.9 kDa, and the supplier reports an observed band at about 32 kDa.

Kallikrein-1 (KLK1), or tissue kallikrein, is a serine protease secreted by the kidney, pancreas and salivary glands. It cleaves low-molecular-weight kininogen to release Lys-bradykinin (kallidin), a peptide that acts on bradykinin receptors to promote vasodilation and local blood flow. KLK1 is studied in blood pressure regulation, kidney function and tissue protection.

Observed MW 32 kDa
Storage As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.

Validated Applications

Western blot 0.1–0.5 µg/mL
ELISA (capture antibody) 1–5 µg/mL

Samples with a confirmed band (WB): recombinant human KLK1 protein.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.

Immunogen

Recombinant human Kallikrein 1 fragment (Ile25–Ser262), expressed in E. coli.

Reactivity Notes

The supplier lists reactivity with human. Tissue expression noted by UniProt (human): Isoform 2 is expressed in pancreas, salivary glands, kidney, colon, prostate gland, testis, spleen and the colon adenocarcinoma cell line T84. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL; ELISA (capture antibody) 1–5 µg/mL. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 28.9 kDa; observed band at about 32 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Recombinant human Kallikrein 1 fragment (Ile25–Ser262), expressed in E. coli.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
