Product Overview
This rabbit polyclonal antibody recognizes voltage-gated inwardly rectifying potassium channel KCNH2 in human, mouse and rat samples and is validated for Western blot and ELISA. The predicted molecular weight is 126.7 kDa, and the supplier reports an observed band at about 150 kDa.
hERG (KCNH2, Kv11.1) is the pore-forming subunit of the channel that carries the rapid delayed rectifier current (IKr) in the heart. Its unusual gating, with rapid inactivation and slow closing, lets it conduct most strongly during repolarization of the cardiac action potential. Loss-of-function mutations cause long QT syndrome type 2, and hERG blockade is a routine safety screen in drug development.

Samples with a confirmed band (WB): human K562 cells, human U2OS cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.

Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places voltage-gated inwardly rectifying potassium channel KCNH2 in the cell membrane. Tissue expression noted by UniProt (human): Highly expressed in heart and brain. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
