Rabbit Monoclonal

Product Overview

Clone EI-11 is a rabbit monoclonal antibody that recognizes kinesin-like protein KIFC1 in human samples and is validated for Western blot, IHC, ICC/IF and immunoprecipitation. The predicted molecular weight is 73.7 kDa, and the supplier reports an observed band at about 74 kDa.

KIFC1 (HSET) is a kinesin-14 family motor that moves toward microtubule minus ends. It crosslinks and focuses microtubules at spindle poles, and it is especially important for clustering extra centrosomes into a bipolar spindle in cancer cells. KIFC1 is studied in mitosis and as a possible target for cancers with centrosome amplification.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200
Immunoprecipitation 1:20

Samples with a confirmed band (WB): human HeLa cells, human Jurkat cells, human HepG2 cells, human MCF-7 cells, human SH-SY5Y cells, human U2OS cells, human THP-1 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (IHC): human thyroid carcinoma tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Immunogen

Synthetic peptide based on the sequence of human KIFC1.

Reactivity Notes

The supplier lists reactivity with human. UniProt places kinesin-like protein KIFC1 in the nucleus, cytoplasm and early endosome. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200; Immunoprecipitation 1:20. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.Has this antibody been tested on feline samples?
A.Reactivity with feline samples has not been tested by the supplier. Aligning the immunogen sequence with the feline ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 73.7 kDa; observed band at about 74 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human KIFC1.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
