Product Overview
This rabbit polyclonal antibody targets importin subunit alpha-1 (gene KPNA2) in human, mouse and rat samples and is validated for Western blot, IHC, flow cytometry and ELISA. The predicted molecular weight is 57.9 kDa, and the supplier reports an observed band at about 58 kDa.
Importin α1 (KPNA2) is an adapter that binds classical nuclear localization signals on cargo proteins and links them to importin β1 for transport through the nuclear pore, with RanGTP triggering release inside the nucleus. KPNA2 is highly expressed in proliferating cells and is often elevated in tumors, making it a subject of cancer research.

|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human, Rat) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human A549 cells, human U251 cells, human U2OS cells, human Daudi cells, human MCF-7 cells, human T-47D cells, human Caco-2 cells, rat testis tissue, mouse testis tissue, mouse RAW264.7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): rat testis tissue, human adenocarcinoma of the right colon tissue, human lymphoma tissue, human lung cancer tissue, human tonsil tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with a confirmed signal (flow cytometry): U87 cells.
Immunogen
Recombinant human KPNA2 fragment (Arg51–Phe529), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places importin subunit alpha-1 in the cytoplasm, nucleus and Golgi apparatus membrane. Tissue expression noted by UniProt (human): Expressed ubiquitously. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
