Product Overview
This rabbit polyclonal antibody targets lethal(3)malignant brain tumor-like protein 2 (gene L3MBTL2) in human samples and is validated for Western blot, IHC, ICC/IF and ELISA. The predicted molecular weight is 79.1 kDa, and the supplier reports an observed band at about 79 kDa.
L3MBTL2 is a Polycomb group-related protein that contains MBT repeats able to bind methylated lysines on histones. It is a component of a noncanonical Polycomb repressive complex 1 (PRC1.6) and the E2F6.com complex, which help keep certain genes, including some involved in cell cycle entry and germ cell development, silent. L3MBTL2 is needed for early embryonic development and is studied in chromatin repression.

Samples with a confirmed band (WB): human 293T cells, human HeLa cells, human MOLT-4 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human colorectal adenocarcinoma tissue, human esophageal squamous carcinoma tissue, human liver cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): U2OS cells.

The supplier lists reactivity with human. UniProt places lethal(3)malignant brain tumor-like protein 2 in the nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
