Product Overview
This rabbit polyclonal antibody recognizes linker for activation of T-cells family member 1 (gene LAT) in human samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 27.9 kDa, and the supplier reports an observed band at about 37 kDa.
LAT (linker for activation of T cells) is a transmembrane adaptor that becomes heavily tyrosine-phosphorylated after T-cell receptor engagement. Phosphorylated LAT recruits PLC-γ1, GRB2, GADS and other signaling proteins, linking the receptor to calcium mobilization, PKC and MAPK activation. It is essential for T-cell development and also functions in mast cell and NK cell receptor signaling.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| Flow cytometry (fixed cells) | 1–3 µg /1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human HEL cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): HEL cells.
Immunogen
Recombinant human LAT fragment (His28–Glu235), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 61% and 59.3%, respectively.
Reactivity Notes
The supplier lists reactivity with human. UniProt places linker for activation of T-cells family member 1 in the cell membrane. Tissue expression noted by UniProt (human): Expressed in thymus, T-cells, NK cells, mast cells and, at lower levels, in spleen. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
