Product Overview
This rabbit polyclonal antibody detects linker for activation of T-cells family member 1 (gene Lat) in mouse and rat samples and is validated for Western blot, IHC, flow cytometry and ELISA. The predicted molecular weight is 26.0 kDa, and the supplier reports an observed band at about 38 kDa.
LAT (linker for activation of T cells) is a transmembrane adaptor that becomes heavily tyrosine-phosphorylated after T-cell receptor engagement. Phosphorylated LAT recruits PLC-γ1, GRB2, GADS and other signaling proteins, linking the receptor to calcium mobilization, PKC and MAPK activation. It is essential for T-cell development and also functions in mast cell and NK cell receptor signaling.

Validated Applications
| Western blot | 0.1–0.25 µg/mL (Mouse) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Mouse, Rat) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Mouse) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): mouse thymus tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse intestinal lymph nodes tissue, mouse spleen tissue, rat intestinal lymph nodes tissue, rat lymph node tissue, rat spleen tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with a confirmed signal (flow cytometry): RAW264.7 cells.
Immunogen
Recombinant mouse Lat fragment (Arg29–Glu217), expressed in E. coli. Sequence identity with the human and rat orthologs: 68.4% and 93.1%, respectively.
Reactivity Notes
The supplier lists reactivity with mouse and rat. UniProt places linker for activation of T-cells family member 1 in the cell membrane. Tissue expression noted by UniProt (mouse): Expressed in T-cells and mast cells. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
