Product Overview
This rabbit polyclonal antibody targets D-lactate dehydrogenase, mitochondrial (gene LDHD) in human, mouse and rat samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 54.9 kDa, and the supplier reports an observed band at about 45–50 kDa.
D-lactate dehydrogenase (LDHD) is a mitochondrial enzyme that oxidizes D-lactate and related D-2-hydroxyacids to their keto acids. D-lactate comes mainly from methylglyoxal metabolism and gut bacteria, and LDHD prevents its buildup. The enzyme is studied in mitochondrial metabolism and in rare inherited D-lactic acid disorders.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human hepatocellular carcinoma paracancerous tissue (HCCP), rat kidney tissue, mouse kidney tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): K562 cells.
Immunogen
Recombinant human LDHD fragment (Arg56–Gln498), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places D-lactate dehydrogenase, mitochondrial in the mitochondrion. Tissue expression noted by UniProt (human): Expressed moderately in heart and liver and at lower levels in skeletal muscle and kidney. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
