Product Overview
This rabbit polyclonal antibody recognizes RNA polymerase-associated protein LEO1 in human, mouse and rat samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 75.4 kDa, and the supplier reports an observed band at about 105 kDa.
LEO1 is a subunit of the PAF1 complex (PAF1C), which binds RNA polymerase II and supports transcription elongation and associated histone modifications. PAF1C helps express developmental genes such as Hox and Wnt targets and helps maintain embryonic stem cell pluripotency. LEO1 is studied in transcription regulation.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human HeLa cells, human MCF-7 cells, human HepG2 cells, human A549 cells, human RT4 cells, rat brain tissue, mouse C2C12 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): HeLa cells.
Samples with a confirmed signal (flow cytometry): Jurkat cells.
Immunogen
Recombinant human LEO1 fragment (Glu355–Arg593), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places RNA polymerase-associated protein LEO1 in the nucleus. Tissue expression noted by UniProt (human): Highly expressed in skeletal muscle and heart. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
