Product Overview
Clone ABDO-12 is a rabbit monoclonal antibody that recognizes leptin receptor (gene LEPR) in human, mouse and rat samples and is validated for Western blot. The predicted molecular weight is 132.5 kDa, and the supplier reports an observed band at about 150 kDa.
The leptin receptor (LEPR, OB-R) is a class I cytokine receptor that binds the adipocyte hormone leptin. Its long isoform, highly expressed in the hypothalamus, activates JAK2–STAT3 signaling to suppress appetite and increase energy expenditure. Loss of LEPR causes severe obesity, as in db/db mice, and the receptor is central to obesity and metabolic research.

Samples with a confirmed band (WB): human SH-SY5Y cells, human K562 cells, human HEL cells, rat kidney tissue, rat spleen tissue, mouse kidney tissue, mouse spleen tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.

Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places leptin receptor in the cell membrane, basolateral cell membrane and SUBCELLULAR LOCATION:. Tissue expression noted by UniProt (human): Isoform A is expressed in fetal liver and in hematopoietic tissues and choroid plexus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
