  • Antibody format: Monoclonal; clone ABDO-12; isotype Rabbit IgG.
  • Host: Rabbit.
  • Species reactivity: Human,Mouse,Rat (confirm in your model system with appropriate controls).

This description is intended to help interpret the antibody design and the biological context of the target using the fields provided in the catalog record, alongside general experimental considerations.

Biological background

LEPR (protein: T-cell surface glycoprotein CD4) is a commonly studied target in molecular and cellular biology. Functional context (as provided): Receptor for obesity factor (leptin). On ligand binding, mediates signaling through JAK2/STAT3. Involved in the regulation of fat metabolism and, in a hematopoietic pathway, required for normal lymphopoiesis. May play a role in reproduction. Can also mediate the ERK/FOS signaling pathway (By similarity). . Reported cellular localization context: Cell membrane; Single-pass type I membrane protein. Tissue expression notes (as provided): Isoform A is expressed in fetal liver and in hematopoietic tissues and choroid plexus. In adults highest expression in heart, liver, small intestine, prostate and ovary. Low level in lung and kidney. Isoform B is highly expressed in hypothalamus.

Research relevance and current trends

  • Research context keywords from the source record include: Collagen,Cytoskeleton/ECM,ECM Proteins,Extracellular Matrix,Signal Transduction,Structures.
  • Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
  • Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.

Common research applications

  • Western blotting (WB): assess relative target abundance across samples, treatments, or time-points.

Workflow ideas (metafield): Validate LEPR antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect LEPR expression by Western blot in cell or tissue lysates, Compare relative LEPR levels across experimental conditions (dose/time-course) using antibody-based readouts

Notes for experimental interpretation

  • Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
  • Apparent molecular weight may vary by sample type and processing (observed MW: 23 kDa; calculated MW: 132494 MW).
  • Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.

Additional product details (from the source record)

  • Molecular weight (observed): 23 kDa
  • Cellular localization (provided): Cell membrane; Single-pass type I membrane protein.
  • Tissue details (provided): Isoform A is expressed in fetal liver and in hematopoietic tissues and choroid plexus. In adults highest expression in heart, liver, small intestine, prostate and ovary. Low level in lung and kidney. Isoform B is highly expressed in hypothalamus.

Does M00350 anti-Leptin Receptor Rabbit Monoclonal antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?
It shows on the product datasheet, M00350 anti-Leptin Receptor Rabbit Monoclonal antibody as been tested on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
