Product Overview
Clone 20L63 is a rabbit monoclonal antibody that recognizes LIM domain kinase 1 (gene LIMK1) in human, mouse and rat samples and is validated for Western blot. The predicted molecular weight is 72.6 kDa, and the supplier reports an observed band at about 73 kDa.
LIM kinase 1 (LIMK1) is a serine/threonine kinase that controls actin dynamics downstream of Rho family GTPases. After activation by ROCK or PAK, it phosphorylates and inactivates cofilin, which stabilizes actin filaments. LIMK1 is studied in cell migration, dendritic spine and synapse biology, and it is one of the genes deleted in Williams syndrome.

Validated Applications
| Western blot | 1:500–1:2000 |
|---|
Samples with a confirmed band (WB): human U87 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Immunogen
Synthetic peptide based on the sequence of human LIM Kinase 1.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places LIM domain kinase 1 in the cytoplasm, nucleus and cell projection. Tissue expression noted by UniProt (human): Highest expression in both adult and fetal nervous system. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
