Product Overview

Clone EAA-12 is a rabbit monoclonal antibody that recognizes protein lin-28 homolog B (gene LIN28B) in human samples and is validated for Western blot, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 27.1 kDa, and the supplier reports an observed band at about 35 kDa.

LIN28B is an RNA-binding protein and paralog of LIN28A that suppresses production of let-7 microRNAs. It binds primary let-7 transcripts in the nucleus and keeps them from being processed, which supports stem cell self-renewal and fetal growth programs. LIN28B is studied in development, fetal hematopoiesis, puberty timing and cancers such as neuroblastoma.

Validated Applications

Western blot 1:500–1:2000
Immunocytochemistry / Immunofluorescence 1:50–1:200
Immunoprecipitation 1:20
Flow cytometry 1:20

Samples with a confirmed band (WB): human K562 cells, human 293T cells, human HepG2 cells, human HUH-7 cells, human HEL cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.

Samples with confirmed staining (ICC/IF): MCF-7 cells.

Immunogen

Synthetic peptide based on the sequence of human lin28B.

Reactivity Notes

The supplier lists reactivity with human. UniProt places protein lin-28 homolog B in the nucleus and cytoplasm. Tissue expression noted by UniProt (human): Expressed at high levels in the placenta and, at mucher lower, in testis and fetal liver. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunocytochemistry / Immunofluorescence 1:50–1:200; Flow cytometry 1:20; Immunoprecipitation 1:20. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 27.1 kDa; observed band at about 35 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human lin28B.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
