Product Overview
This rabbit polyclonal antibody recognizes E3 ubiquitin-protein ligase TRIM71 in human, mouse and rat samples and is validated for Western blot, IHC, flow cytometry, immunoprecipitation and ELISA. The predicted molecular weight is 93.4 kDa, and the supplier reports an observed band at about 93 kDa.
TRIM71 (LIN-41) is a tripartite motif E3 ubiquitin ligase and RNA-binding protein that is highly expressed in embryonic stem cells and early development. It represses target mRNAs, in part together with the microRNA machinery, and supports stem cell self-renewal. It is itself a classic target of the let-7 microRNA and is studied in developmental timing and neural tube formation.

| Western blot | 0.1–0.25 µg/mL (Human) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human, Mouse, Rat) |
| Immunoprecipitation | 0.5–2 µg/mL (Human) |

Samples with a confirmed band (WB): human HepG2 cells, human Caco-2 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human lung cancer tissue, human endometrial adenocarcinoma tissue, human prostate adenocarcinoma tissue, mouse testis tissue, rat testis tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with a confirmed result (IP): HepG2 cells.
Samples with a confirmed signal (flow cytometry): HEL cells.
Immunogen
Recombinant human LIN41/TRIM71 fragment (Glu85–Phe868), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places E3 ubiquitin-protein ligase TRIM71 in the cytoplasm. Tissue expression noted by UniProt (human): Specifically expressed in testis. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
