Product Overview
This rabbit polyclonal antibody detects protein-lysine 6-oxidase (gene LOX) in human, mouse and rat samples and is validated for ELISA and Western blot. The predicted molecular weight is 46.9 kDa, and the supplier reports an observed band at about 47 kDa.
Lysyl oxidase (LOX) is a copper-dependent secreted enzyme that oxidizes lysine and hydroxylysine residues in collagen and elastin. The resulting aldehydes form covalent cross-links that give connective tissues tensile strength and elasticity, and LOX is needed for the structural integrity of the aorta. LOX is studied in fibrosis, vascular biology and cancer metastasis.

Samples with a confirmed band (WB): rat liver tissue, rat lung tissue, mouse liver tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Synthetic peptide from the middle region of human LOX. It differs from the mouse and rat sequences at one residue.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt describes protein-lysine 6-oxidase as a secreted protein. Tissue expression noted by UniProt (human): Heart, placenta, skeletal muscle, kidney, lung and pancreas. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
