Product Overview
Clone ABEO-12 is a rabbit monoclonal antibody that recognizes protein-lysine 6-oxidase (gene LOX) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 46.9 kDa, and the supplier reports an observed band at about 47 kDa.
Lysyl oxidase (LOX) is a copper-dependent secreted enzyme that oxidizes lysine and hydroxylysine residues in collagen and elastin. The resulting aldehydes form covalent cross-links that give connective tissues tensile strength and elasticity, and LOX is needed for the structural integrity of the aorta. LOX is studied in fibrosis, vascular biology and cancer metastasis.

Samples with a confirmed band (WB): human SiHa cells, rat brain tissue, mouse brain tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.
Samples with confirmed staining (IHC): human colorectal adenocarcinoma tissue, human lung squamous cells carcinoma tissue, human spleen tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): Jurkat cells.
Immunogen
Synthetic peptide based on the sequence of human LOX.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt describes protein-lysine 6-oxidase as a secreted protein. Tissue expression noted by UniProt (human): Heart, placenta, skeletal muscle, kidney, lung and pancreas. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
