Product Overview
This rabbit polyclonal antibody targets leucine-rich repeat, immunoglobulin-like domain and transmembrane domain-containing protein 3 (gene LRIT3) in human, mouse and rat samples and is validated for Western blot, ICC/IF and ELISA. The predicted molecular weight is 74.8 kDa, and the supplier reports an observed band at about 75 kDa.
LRIT3 is a leucine-rich repeat, immunoglobulin-like and transmembrane protein of the retina. It is needed for signal transfer from photoreceptors to ON-bipolar cells and for placing the TRPM1 cation channel at bipolar cell dendrite tips, so loss of LRIT3 disrupts ON-pathway transmission; it has also been linked to FGFR trafficking, and it is studied in retinal synapse biology and congenital stationary night blindness.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human 293T cells, human Jurkat cells, rat kidney tissue, mouse kidney tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): U2OS cells.
Immunogen
Recombinant human LRIT3 fragment (Glu50–Arg573), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places leucine-rich repeat, immunoglobulin-like domain and transmembrane domain-containing protein 3 in the cell projection, perikaryon and endoplasmic reticulum membrane. Tissue expression noted by UniProt (human): Detected in the outer plexiform layer (OPL) of the retina where it localizes to ON-bipolar cells (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
