Product Overview
Clone DIC-9 is a rabbit monoclonal antibody that recognizes cation-independent mannose-6-phosphate receptor (gene IGF2R) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 274.4 kDa, and the supplier reports an observed band at about 300 kDa.
IGF2R, the cation-independent mannose-6-phosphate receptor, is a large transmembrane receptor that carries newly made lysosomal enzymes tagged with mannose-6-phosphate from the Golgi to endosomes and lysosomes. It also binds IGF-2 at the cell surface and directs it for degradation, thereby limiting IGF-2 signaling.

Samples with a confirmed band (WB): human HepG2 cells, human THP-1 cells, human U251 cells, human SiHa cells, rat heart tissue, rat lung tissue, mouse heart tissue, mouse lung tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 8% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human colon tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Immunogen
Synthetic peptide based on the sequence of human M6PR.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places cation-independent mannose-6-phosphate receptor in the Golgi apparatus membrane and endosome membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
