Product Overview
This rabbit polyclonal antibody recognizes C-C motif chemokine 4 (gene Ccl4) in rat samples and is validated for Western blot and IHC. The predicted molecular weight is 10.2 kDa, and the supplier reports an observed band at about 11 kDa.
CCL4, also known as MIP-1β, is a CC chemokine produced by activated monocytes, macrophages and CD8+ T cells. It acts mainly through CCR5 to attract immune cells to sites of inflammation, and by occupying CCR5 it can block entry of CCR5-tropic HIV-1, which makes it a frequent target in immunology and HIV research.

Validated Applications
| Western blot | 0.1–0.5 µg/mL (Rat) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Rat) |
Samples with a confirmed band (WB): recombinant rat CCL4 protein.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): rat spleen.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Recombinant rat CCL4 fragment (Ala24–Asn92), expressed in E. coli. Sequence identity with the human and mouse orthologs: 80% and 86%, respectively.
Reactivity Notes
The supplier lists reactivity with rat. UniProt describes C-C motif chemokine 4 as a secreted protein. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
