Overview

This antibody is intended for detection of Mmp12 (Macrophage metalloelastase) in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.

Vendor notes: Boster Bio Anti-Macrophage metalloelastase MMP12 Antibody catalog # RP1089. Tested in ELISA, WB applications. This antibody reacts with Mouse. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: A synthetic peptide corresponding to a sequence at the C-terminus of mouse MMP12, different from the related human sequence by thirteen amino acids, and from the related rat sequence by three amino acids.
  • Molecular weight context: reported MW: 55 kDa; calculated MW: 54971 MW
  • Reactivity: Mouse
  • Applications: ELISA, WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

Macrophage metalloelastase; Macrophage metalloelastase. Matrix metalloproteinase-12 (MMP12), also known as MME or ME, is an enzyme that in humans is encoded by the MMP12 gene. The gene is part of a cluster of MMP genes which localize to chromosome 11q22.2. It is thought that the protein encoded by this gene is cleaved at both ends to yield the active enzyme, but this processing has not been fully described. The enzyme degrades soluble and insoluble elastin. It may play a role in aneurysm formation and studies in mice suggest a role in the development of emphysema. This gene may involved in tissue injury and remodeling. Functional note: May be involved in tissue injury and remodeling. Has significant elastolytic activity. Can accept large and small amino acids at the P1' site, but has a preference for leucine. Aromatic or hydrophobic residues are preferred at the P1 site, with small hydrophobic residues (preferably alanine) occupying P3 (By similarity). . Reported localization: Secreted, extracellular space, extracellular matrix .

Research relevance and current trends

  • Angiogenesis: Researchers commonly examine how Mmp12 (Macrophage metalloelastase) relates to this theme using model systems and orthogonal readouts.
  • Atherosclerosis: Researchers commonly examine how Mmp12 (Macrophage metalloelastase) relates to this theme using model systems and orthogonal readouts.
  • Cancer: Researchers commonly examine how Mmp12 (Macrophage metalloelastase) relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative Mmp12 (Macrophage metalloelastase) levels across conditions; band patterns may reflect isoforms and processing.
  • ELISA-compatible use: when applicable, interpret signal as relative abundance across sample sets with consistent handling and dilution strategy.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.

Our team were well pleased with the WB result of your anti-MMP12 antibody. However we have observed positive staining in amniotic fluid extracellular space using this antibody. Is that expected? Could you tell me where is MMP12 supposed to be expressed?
According to literature, amniotic fluid does express MMP12. Generally MMP12 expresses in secreted, extracellular space, extracellular. Regarding which tissues have MMP12 expression, here are a few articles citing expression in various tissues: Alveolar macrophage, Pubmed ID: 8226919 Esophagus, Pubmed ID: 14702039
