Product Overview

This rabbit polyclonal antibody targets mucosal addressin cell adhesion molecule 1 (gene MADCAM1) in human and rat samples and is validated for Western blot. The predicted molecular weight is 40.2 kDa, and the supplier reports an observed band at about 50 kDa.

Mucosal addressin cell adhesion molecule 1 (MADCAM1) is an immunoglobulin-superfamily adhesion molecule expressed on high endothelial venules of gut-associated lymphoid tissue and intestinal venules. By binding integrin α4β7 and L-selectin it guides lymphocytes into Peyer's patches and the intestinal lamina propria, so it is central to studies of gut immune homing and inflammatory bowel disease.

Validated Applications

Western blot 0.25–0.5 µg/mL (Human, Rat)

Samples with a confirmed band (WB): human MCF-7 cells, rat RH35 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Synthetic peptide from the middle region of human MADCAM1. Sequence identity with the mouse ortholog: 46.2%.

Reactivity Notes

The supplier lists reactivity with human and rat. UniProt places mucosal addressin cell adhesion molecule 1 in the membrane. Tissue expression noted by UniProt (human): Highly expressed on high endothelial venules (HEV) and lamina propia venules found in the small intestine, and to a lesser extent in the colon and spleen. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.25–0.5 µg/mL (Human, Rat). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 40.2 kDa; observed band at about 50 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the middle region of human MADCAM1. Sequence identity with the mouse ortholog: 46.2%.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
