Product Overview
Clone 28M41 is a rabbit monoclonal antibody that recognizes mitogen-activated protein kinase kinase kinase 4 (gene MAP3K4) in human samples and is validated for Western blot and IHC. The predicted molecular weight is 181.7 kDa, and the supplier reports an observed band at about 182 kDa.
MEKK4 (MAP3K4) is a MAP kinase kinase kinase that activates the p38 and JNK stress pathways, but not ERK, by phosphorylating MKK4 and MKK6. It is activated by GADD45 proteins after cellular stress and is also important for neural tube development.

Validated Applications
| Western blot | 1:500–1:2000 |
|---|---|
| Immunohistochemistry | 1:50–1:200 |
Samples with a confirmed band (WB): human U251 cells, human K562 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 8% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1: 500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Immunogen
Synthetic peptide based on the sequence of human MAP3K4.
Reactivity Notes
The supplier lists reactivity with human. UniProt places mitogen-activated protein kinase kinase kinase 4 in the cytoplasm. Tissue expression noted by UniProt (human): Expressed at high levels in heart, placenta, skeletal muscle and pancreas, and at lower levels in other tissues. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
