Product Overview

This rabbit polyclonal antibody detects mitogen-activated protein kinase kinase kinase kinase (gene Map4k1) in rat samples and is validated for Western blot.

HPK1 (hematopoietic progenitor kinase 1, MAP4K1) is a serine/threonine kinase of the germinal-center kinase family expressed mainly in blood cells. It activates the JNK pathway and acts as a negative regulator of T-cell receptor and B-cell receptor signaling, which has made it a target of interest in immuno-oncology research.

Validated Applications

Western blot 0.25–0.5 µg/mL (Rat)

Samples with a confirmed band (WB): rat thymus tissue, rat NRK cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Synthetic peptide from the N-terminal region of rat MAP4K1/HPK1. Sequence identity with the human and mouse orthologs: 88.2% and 100%, respectively.

Reactivity Notes

The supplier lists reactivity with rat. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
