Vendor description (summary): Boster Bio Anti-MAP4K5 Antibody Picoband® catalog # A10186-1.
Biological background
Biological context: Promotes the growth of epithelial cells. May stimulate the phosphorylation of EGFR and mitogen-activated protein kinases.
Expression and localization notes: cellular localization: Membrane ; Single-pass type I membrane protein ., tissue context: Ubiquitous. Highly expressed in brain. ..
Common research applications
- Western blotting (WB): Compare MAP4K5 levels across samples and conditions using appropriate loading and biological controls.
- Immunohistochemistry (IHC): Evaluate spatial distribution of MAP4K5 in tissue sections, considering fixation and antigen retrieval effects.
- Immunofluorescence / ICC: Assess subcellular localization patterns and co-localization with compartment markers in cultured cells.
- Flow cytometry: Quantify MAP4K5-positive populations in single-cell suspensions with appropriate gating and controls.
- ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.
Notes for experimental interpretation
- Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
- Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.
Additional product notes (from provided fields)
- Background: Mitogen-activated protein kinase kinase kinase kinase 5 is an enzyme that in humans is encoded by the MAP4K5 gene. This gene encodes a member of the serine/threonine protein kinase family, that is highly similar to yeast SPS1/STE20 kinase. Yeast SPS1/STE20 functions near the beginning of the MAP kinase signal cascades that is essential for yeast pheromone response. This kinase was shown to activate Jun kinase in mammalian cells, which suggested a role in stress response. Two alternatively spliced transcript variants encoding the same protein have been described for this gene.
- Cross reactivity: No cross-reactivity with other proteins
- Cellular localization: Membrane ; Single-pass type I membrane protein .
- Tissue details: Ubiquitous. Highly expressed in brain. .
