Product Overview

Clone FHH-20 is a rabbit monoclonal antibody that recognizes tryptase alpha/beta-1 (gene TPSAB1) in human, mouse and rat samples and is validated for Western blot, IHC and immunoprecipitation. The predicted molecular weight is 30.5 kDa, and the supplier reports an observed band at about 30 kDa.

Tryptase alpha/beta-1 (TPSAB1) encodes tryptase, the most abundant serine protease stored in mast cell granules and released when mast cells degranulate. Tryptase cleaves extracellular substrates such as fibronectin and may contribute to innate immunity. Serum tryptase is a standard marker of mast cell activation and burden in allergy and mastocytosis research.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunoprecipitation 1:20

Samples with a confirmed band (WB): human tonsil.

Recommended loading (WB): 20–40 µg of total protein per lane.

Samples with confirmed staining (IHC): human kidney tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Immunogen

Synthetic peptide based on the sequence of human mast Cell Tryptase.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt describes tryptase alpha/beta-1 as a secreted protein. Tissue expression noted by UniProt (human): Isoform 1 and isoform 2 are expressed in lung, stomach, spleen, heart and skin. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunoprecipitation 1:20. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Is a carrier-free (BSA-free) version available, for example for conjugation?
A.Yes. A carrier-free formulation without BSA, stabilized with trehalose and/or glycerol, can be prepared on request and is compatible with common conjugation chemistries. Please note this when ordering or contact info@biohippo.com. For storage at −20 °C, keep a cryoprotectant such as glycerol in the buffer rather than PBS alone.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 30.5 kDa; observed band at about 30 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human mast Cell Tryptase.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
