This description is intended to help interpret the antibody design and the biological context of the target using the fields provided in the catalog record, alongside general experimental considerations.

Biological background

MBD2 (protein: Lysosome-associated membrane glycoprotein 2 (Lamp2)) is a commonly studied target in molecular and cellular biology. Functional context (as provided): Regulatory component of the cyclin D3-CDK4 (DC) complex that phosphorylates and inhibits members of the retinoblastoma (RB) protein family including RB1 and regulates the cell-cycle during G (1)/S transition. Phosphorylation of RB1 allows dissociation of the transcription factor E2F from the RB/E2F complex and the subsequent transcription of E2F target genes which are responsible for the progression through the G (1) phase. Hypophosphorylates RB1 in early G (1) phase. Cyclin D-CDK4 complexes are major integrators of various mitogenenic and antimitogenic signals. Also substrate for SMAD3, phosphorylating SMAD3 in a cell-cycle-dependent manner and repressing its transcriptional activity. Component of the ternary complex, cyclin D3/CDK4/CDKN1B, required for nuclear translocation and activity of the cyclin D-CDK4 complex. . Reported cellular localization context: Nucleus . Cytoplasm . Membrane . Cyclin D-CDK4 complexes accumulate at the nuclear membrane and are then translocated to the nucleus through interaction with KIP/CIP family members. . Tissue expression notes (as provided): Neutrophils.

Research relevance and current trends

  • Research context keywords from the source record include: 2339,Epigenetics and Nuclear Signaling.
  • Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
  • Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.

Common research applications

  • Western blotting (WB): assess relative target abundance across samples, treatments, or time-points.
  • Immunohistochemistry (IHC): evaluate spatial distribution of target-positive staining in tissue architecture.
  • Immunofluorescence/ICC (IF/ICC): visualize subcellular localization patterns and cell-to-cell heterogeneity.
  • Flow cytometry: quantify target-positive populations and compare shifts in marker distributions.
  • Immunoprecipitation (IP): enrich target complexes for downstream immunoblot or interaction analyses.

Workflow ideas (metafield): Validate MBD2 antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect MBD2 expression by Western blot in cell or tissue lysates, Detect MBD2 in FFPE tissue sections by immunohistochemistry, Localize MBD2 by immunofluorescence/immunocytochemistry in cultured cells, Quantify MBD2-positive cells by flow cytometry in single-cell suspensions, Enrich MBD2 by immunoprecipitation from lysates for downstream analysis

Notes for experimental interpretation

  • Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
  • Apparent molecular weight may vary by sample type and processing (observed MW: 56 kDa; calculated MW: 32520 MW).
  • Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.

Additional product details (from the source record)

  • Molecular weight (observed): 56 kDa
  • Cellular localization (provided): Nucleus . Cytoplasm . Membrane . Cyclin D-CDK4 complexes accumulate at the nuclear membrane and are then translocated to the nucleus through interaction with KIP/CIP family members. .
  • Tissue details (provided): Neutrophils.

Would M01746 anti-MBD2 Monoclonal antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?
As indicated on the product datasheet, M01746 anti-MBD2 Monoclonal antibody as been validated on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
