Product Overview

Clone AFEE-13 is a rabbit monoclonal antibody that recognizes methyl-CpG-binding domain protein 3 (gene MBD3) in human, mouse and rat samples and is validated for Western blot, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 32.8 kDa, and the supplier reports an observed band at about 35 kDa.

MBD3 is a core subunit of the NuRD (nucleosome remodeling and deacetylase) complex. Unlike MBD1 and MBD2, it binds methylated DNA only weakly, but it is essential for NuRD assembly and function. MBD3 is studied in embryonic development, stem cell pluripotency and reprogramming, and chromatin-based gene regulation.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places methyl-CpG-binding domain protein 3 in the nucleus and chromosome. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunocytochemistry / Immunofluorescence 1:50–1:200; Flow cytometry 1:50; Immunoprecipitation 1:50. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on zebrafish samples?
A.Reactivity with zebrafish samples has not been tested by the supplier. Aligning the immunogen sequence with the zebrafish ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 32.8 kDa; observed band at about 35 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human MBD3 Does not bind DNA by itself.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
