Product Overview

Clone 26M32 is a rabbit monoclonal antibody that recognizes methyl-CpG-binding domain protein 3 (gene MBD3) in human, mouse and rat samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 32.8 kDa; the supplier reports observed bands at 33, 34, 36 kDa.

MBD3 is a core subunit of the NuRD (nucleosome remodeling and deacetylase) complex. Unlike MBD1 and MBD2, it binds methylated DNA only weakly, but it is essential for NuRD assembly and function. MBD3 is studied in embryonic development, stem cell pluripotency and reprogramming, and chromatin-based gene regulation.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200

Samples with a confirmed band (WB): human HeLa cells, human RT4 cells, human U251 cells, human A549 cells, rat brain tissue, rat C6 cells, mouse brain tissue, mouse Neuro-2a cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:1000, ECL detection.

Samples with confirmed staining (ICC/IF): HeLa cells.

Immunogen

Synthetic peptide based on the sequence of human MBD3.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places methyl-CpG-binding domain protein 3 in the nucleus and chromosome. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
