Product Overview
This rabbit polyclonal antibody detects methyl-CpG-binding domain protein 4 (gene Mbd4) in mouse and rat samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 62.6 kDa, and the supplier reports an observed band at about 66 kDa.
MBD4 is a DNA glycosylase that combines a methyl-CpG-binding domain with a repair enzyme domain. It removes thymine or uracil mispaired with guanine, especially at CpG sites, correcting damage that arises from deamination of methylated or unmethylated cytosine. It is studied in DNA repair, mutation rates at CpG sites and cancer.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Mouse, Rat) |
|---|---|
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Mouse) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): rat liver tissue, mouse liver tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): C2C12 cells.
Immunogen
Recombinant mouse Mbd4 fragment (Arg238–Asp534), expressed in E. coli. Sequence identity with the human ortholog: 69.7%.
Reactivity Notes
The supplier lists reactivity with mouse and rat. UniProt places methyl-CpG-binding domain protein 4 in the nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
