Product Overview
This rabbit polyclonal antibody targets induced myeloid leukemia cell differentiation protein Mcl-1 (gene MCL1) in human, mouse and rat samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 37.3 kDa, and the supplier reports an observed band at about 40 kDa.
MCL-1 is an anti-apoptotic member of the BCL-2 family that sits on the mitochondrial outer membrane. It binds and neutralizes pro-apoptotic proteins such as BAK and BIM, preventing mitochondrial outer membrane permeabilization. MCL-1 turns over rapidly, and because many cancers depend on it for survival, it is studied as a cancer target and in cell death regulation.

Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): PC3 cells.
Samples with a confirmed signal (flow cytometry): RT4 cells.
Immunogen
Recombinant human MCL1 fragment (Glu109–Glu325), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places induced myeloid leukemia cell differentiation protein Mcl-1 in the membrane, cytoplasm, mitochondrion and nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
