Product Overview

This rabbit polyclonal antibody recognizes induced myeloid leukemia cell differentiation protein Mcl-1 (gene MCL1) in human samples and is validated for Western blot. The predicted molecular weight is 37.3 kDa, and the supplier reports an observed band at about 37 kDa.

MCL-1 is an anti-apoptotic member of the BCL-2 family that sits on the mitochondrial outer membrane. It binds and neutralizes pro-apoptotic proteins such as BAK and BIM, preventing mitochondrial outer membrane permeabilization. MCL-1 turns over rapidly, and because many cancers depend on it for survival, it is studied as a cancer target and in cell death regulation.

Validated Applications

Western blot 0.1–0.5 µg/mL (Human)

Samples with a confirmed band (WB): HeLa cells, MCF-7 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Immunogen

Synthetic peptide from the C-terminal region of human MCL1. It differs from the mouse sequence at one residue.

Reactivity Notes

The supplier lists reactivity with human. UniProt places induced myeloid leukemia cell differentiation protein Mcl-1 in the membrane, cytoplasm, mitochondrion and nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide and thimerosal (a mercury compound) as preservatives. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 37.3 kDa; observed band at about 37 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the C-terminal region of human MCL1. It differs from the mouse sequence at one residue.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
