Product Overview
This rabbit polyclonal antibody targets DNA replication licensing factor MCM4 in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 96.6 kDa, and the supplier reports an observed band at about 97 kDa.
MCM4 is a subunit of the MCM2-7 complex, the core of the replicative helicase that unwinds DNA at replication forks. The complex is loaded at origins before S phase and activated to start DNA synthesis. MCM4 is studied in DNA replication licensing, genome stability and cell proliferation.

Samples with a confirmed band (WB): human HeLa cells, human U2OS cells, human MCF-7 cells, human HEK293 cells, human Jurkat cells, human HL-60 cells, rat liver tissue, mouse NIH/3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse colon tissue, rat colon tissue, human breast cancer tissue, human gallbladder adenocarcinoma tissue, human rectal cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): A431 cells.
Samples with a confirmed signal (flow cytometry): A549 cells.
Immunogen
Recombinant human MCM4 fragment (Met361–Ala677), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places DNA replication licensing factor MCM4 in the nucleus and chromosome. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
