Product Overview
This rabbit polyclonal antibody detects DNA helicase MCM8 in human samples and is validated for Western blot. The predicted molecular weight is 93.7 kDa, and the supplier reports an observed band at about 94 kDa.
MCM8 is a helicase related to the replicative MCM proteins but not part of the MCM2-7 complex. Together with MCM9, it forms a helicase that supports homologous recombination repair of DNA double-strand breaks and interstrand cross-links, partly by helping the MRN complex resect DNA ends. Mutations in MCM8 are linked to primary ovarian insufficiency and infertility.

Samples with a confirmed band (WB): human 293T cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Synthetic peptide from the C-terminal region of human MCM8. It differs from the mouse and rat sequences at one residue.
Reactivity Notes
The supplier lists reactivity with human. UniProt places DNA helicase MCM8 in the nucleus, chromosome, cytoplasm and mitochondrion matrix. Tissue expression noted by UniProt (human): Highest levels in placenta, lung and pancreas. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
