Product Overview
Clone 20S76 is a rabbit monoclonal antibody that recognizes monocarboxylate transporter 1 (gene SLC16A1) in human samples and is validated for Western blot and IHC. The predicted molecular weight is 53.9 kDa, and the supplier reports an observed band at about 50 kDa.
Monocarboxylate transporter 1 (MCT1, SLC16A1) moves lactate, pyruvate and ketone bodies across the plasma membrane together with a proton. Because the direction of transport follows substrate and proton gradients, MCT1 supports both lactate export from glycolytic cells and lactate uptake as a fuel, and it is widely studied in tumor metabolism and exercise physiology.

Validated Applications
| Western blot | 1:500–1:2000 |
|---|---|
| Immunohistochemistry | 1:50–1:200 |
Samples with a confirmed band (WB): human K562 cells, human A431 cells, human HepG2 cells, human 293T cells, human PC-3 cells, human SIHA cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human breast cancer tissue, human cervical cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide based on the sequence of human monocarboxylic acid transporter 1.
Reactivity Notes
The supplier lists reactivity with human. UniProt places monocarboxylate transporter 1 in the cell membrane, basolateral cell membrane and apical cell membrane. Tissue expression noted by UniProt (human): Widely expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
