Product Overview
This rabbit polyclonal antibody detects E3 ubiquitin-protein ligase Mdm2 in human, mouse and rat samples and is validated for Western blot. The predicted molecular weight is 55.2 kDa, and the supplier reports an observed band at about 90 kDa.
MDM2 is an E3 ubiquitin ligase that is the principal negative regulator of the tumor suppressor p53. It binds p53, blocks its transcriptional activity and targets it for proteasomal degradation, while p53 in turn induces MDM2, forming a feedback loop. MDM2 is amplified in some cancers and is widely studied in p53 signaling and anticancer drug development.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse, Rat) |
|---|
Samples with a confirmed band (WB): human U-87MG cells, rat PC-12 cells, rat RH-35 cells, mouse HEPA1–6 cells, mouse NTH/3T3 cells, mouse ANA-1 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Immunogen
Synthetic peptide from the N-terminal region of human MDM2. The peptide sequence is conserved in mouse and rat.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places E3 ubiquitin-protein ligase Mdm2 in the nucleus and cytoplasm. Tissue expression noted by UniProt (human): Ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
