Product Overview
This rabbit polyclonal antibody recognizes mediator of RNA polymerase II transcription subunit 17 (gene MED17) in human samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 72.9 kDa, and the supplier reports an observed band at about 73 kDa.
MED17 is a core subunit of the head module of the Mediator complex, which contacts RNA polymerase II and helps assemble the preinitiation complex at promoters. It is essential for Mediator function in transcription of most protein-coding genes. Mutations in MED17 are linked to a severe neurodevelopmental disorder, and it is studied in transcription mechanisms.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human Jurkat cells, human Caco-2 cells, human SIHA cells, human K562 cells, human HeLa cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): U251 cells.
Immunogen
Recombinant human MED17 fragment (Val4–Glu561), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places mediator of RNA polymerase II transcription subunit 17 in the nucleus. Tissue expression noted by UniProt (human): Ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
