Product Overview
This rabbit polyclonal antibody detects mediator of RNA polymerase II transcription subunit 4 (gene MED4) in human, mouse and rat samples and is validated for ELISA, IHC and Western blot. The predicted molecular weight is 29.7 kDa, and the supplier reports an observed band at about 34 kDa.
MED4 is a subunit of the middle module of the Mediator complex, the coactivator that connects gene-specific transcription factors with RNA polymerase II. The middle module links the head and tail and contributes to Mediator′s structural stability. MED4 is studied in transcription and Mediator structure.

Samples with a confirmed band (WB): human Jurkat cells, human THP-1 cells, human HepG2 cells, human HEL cells, rat brain tissue, rat PC-12 cells, mouse brain tissue, mouse Ana-1 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse brain tissue, mouse small intestine tissue, rat brain tissue, rat small intestine tissue, human mammary cancer tissue, human rectal cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).
Immunogen
Recombinant human MED4 fragment (Arg27–Lys140), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places mediator of RNA polymerase II transcription subunit 4 in the nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
