Product Overview
This rabbit polyclonal antibody recognizes myocyte-specific enhancer factor 2A (gene MEF2A) in human, mouse and rat samples and is validated for ICC/IF, IHC and Western blot. The predicted molecular weight is 54.8 kDa, and the supplier reports an observed band at about 56 kDa.
MEF2A is a member of the myocyte enhancer factor 2 family of MADS-box transcription factors. It binds A/T-rich MEF2 sites in the control regions of many muscle-specific and growth factor-responsive genes and cooperates with myogenic factors during muscle differentiation. MEF2A activity is regulated by MAP kinases and class II HDACs, and it is studied in muscle, heart and neuronal biology.

Validated Applications
| Western blot | 0.1–0.5 µg/mL (Human, Rat) |
|---|---|
| IHC (paraffin sections) | 0.5–1 µg/mL (Human, Mouse, Rat) |
| Immunocytochemistry / Immunofluorescence | 2 µg/mL (Human) |
Samples with a confirmed band (WB): rat thymus tissue, HeLa cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse brain tissue, rat brain tissue, human glioma tissue, human meningeoma tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): U2OS cells.
Immunogen
Synthetic peptide from the C-terminal region of human MEF2A. It differs from the mouse sequence at one residue and from the rat sequence at two residues.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places myocyte-specific enhancer factor 2A in the nucleus. Tissue expression noted by UniProt (human): Isoform MEF2 and isoform MEFA are expressed only in skeletal and cardiac muscle and in the brain. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
