Product Overview
This rabbit polyclonal antibody detects pyrin (gene MEFV) in human and rat samples and is validated for Western blot. The predicted molecular weight is 86.4 kDa, and the supplier reports an observed band at about 86 kDa.
Pyrin (encoded by MEFV) is an innate immune sensor expressed mainly in neutrophils and monocytes. It detects inactivation of RhoA GTPases, for example by bacterial toxins, and then assembles an inflammasome that activates caspase-1 and releases IL-1β. Mutations in MEFV cause familial Mediterranean fever, making pyrin central to research on autoinflammatory disease.

Samples with a confirmed band (WB): rat spleen tissue, rat lung tissue, HEPA cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Reactivity Notes
The supplier lists reactivity with human and rat. Tissue expression noted by UniProt (human): Expressed in peripheral blood leukocytes, particularly in mature granulocytes and to a lesser extent in monocytes but not in lymphocytes. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
