Product Overview

Clone ACDO-13 is a rabbit monoclonal antibody that recognizes homeobox protein Meis1 in human, mouse and rat samples and is validated for Western blot, immunoprecipitation and flow cytometry. The predicted molecular weight is 43.0 kDa; the supplier reports observed bands at 45,53 kDa.

MEIS1 is a TALE-class homeodomain transcription factor that partners with PBX and HOX proteins to regulate target genes. It is essential for hematopoietic stem cell maintenance, megakaryocyte and vascular development, and heart development. Together with HOXA9, MEIS1 drives certain acute myeloid leukemias, especially MLL-rearranged forms, and it is studied in development and leukemia.

Validated Applications

Western blot 1:500–1:2000
Immunoprecipitation 1:20
Flow cytometry 1:20

Samples with a confirmed band (WB): human K562 cells, mouse pancrease tissue, human SH-SY5Y cells, human HeLa cells, human HEL cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Synthetic peptide based on the sequence of human MEIS1.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places homeobox protein Meis1 in the nucleus. Tissue expression noted by UniProt (human): Expressed at low level in normal immunohepatopoietic tissues, including the fetal liver. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Flow cytometry 1:20; Immunoprecipitation 1:20. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 43.0 kDa; observed bands at about 45 and 53 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human MEIS1.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
