Product Overview
Clone HBI-13 is a rabbit monoclonal antibody that recognizes dual specificity mitogen-activated protein kinase kinase 2 (gene MAP2K2) in human and mouse samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 44.4 kDa, and the supplier reports an observed band at about 44 kDa.
MEK2 (MAP2K2) is a dual-specificity kinase closely related to MEK1. Activated by RAF kinases, it phosphorylates a threonine and a tyrosine in the activation loop of ERK1 and ERK2 to switch on the ERK pathway, and it is studied in growth factor signaling, RASopathies and cancer.

Samples with a confirmed band (WB): human MCF-7 cells, human HepG2 cells, human U251 cells, human K562 cells, mouse skeletal muscle tissue, mouse brain tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:1000, ECL detection.
Samples with confirmed staining (IHC): human stomach cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with a confirmed result (IP): K562 cells.
Immunogen
Synthetic peptide based on the sequence of human MEK2.
Reactivity Notes
The supplier lists reactivity with human and mouse. UniProt places dual specificity mitogen-activated protein kinase kinase 2 in the cytoplasm and membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
