Product Overview

This rabbit polyclonal antibody targets dual specificity mitogen-activated protein kinase kinase 6 (gene MAP2K6) in human and rat samples and is validated for Western blot. The predicted molecular weight is 37.5 kDa, and the supplier reports an observed band at about 38 kDa.

MKK6 (MAP2K6) is a dual-specificity kinase that, together with MKK3, phosphorylates and activates the p38 MAP kinases. It is switched on by pro-inflammatory cytokines and environmental stress and is studied in inflammation, stress-induced cell death and differentiation.

Validated Applications

Western blot 0.1–0.5 µg/mL (Human, Rat)

Samples with a confirmed band (WB): human HeLa cells, human A549 cells, rat C6 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Recombinant human MEK6 fragment (Ala280–Asp334), expressed in E. coli. Sequence identity with the mouse ortholog: 92.7%.

Reactivity Notes

The supplier lists reactivity with human and rat. UniProt places dual specificity mitogen-activated protein kinase kinase 6 in the nucleus and cytoplasm. Tissue expression noted by UniProt (human): Isoform 2 is only expressed in skeletal muscle. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
