Product Overview

This rabbit polyclonal antibody targets dual specificity mitogen-activated protein kinase kinase 6 (gene MAP2K6) in human, mouse and rat samples and is validated for Western blot. The predicted molecular weight is 37.5 kDa, and the supplier reports an observed band at about 22–24 kDa.

MKK6 (MAP2K6) is a dual-specificity kinase that, together with MKK3, phosphorylates and activates the p38 MAP kinases. It is switched on by pro-inflammatory cytokines and environmental stress and is studied in inflammation, stress-induced cell death and differentiation.

Validated Applications

Western blot 0.1–0.5 µg/mL (Human, Mouse, Rat)

Samples with a confirmed band (WB): rat ovary tissue, rat spleen tissue, rat brain tissue, rat heart tissue, NIH3I3 cells, HeLa cells, JRUKAT cells, MCF-7 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Immunogen

Synthetic peptide from the N-terminal region of human MEK6. The peptide sequence is conserved in mouse. It differs from the rat sequence at one residue.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places dual specificity mitogen-activated protein kinase kinase 6 in the nucleus and cytoplasm. Tissue expression noted by UniProt (human): Isoform 2 is only expressed in skeletal muscle. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide and thimerosal (a mercury compound) as preservatives. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human, Mouse, Rat). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 37.5 kDa; observed bands at about 22 and 24 kDa. The observed band is much smaller than the calculated size, which can indicate a processed form or a shorter isoform; check the supplier images.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the N-terminal region of human MEK6. The peptide sequence is conserved in mouse.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
