Product Overview
Clone 21M06 is a rabbit monoclonal antibody that recognizes N(6)-adenosine-methyltransferase catalytic subunit METTL3 in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 64.5 kDa, and the supplier reports an observed band at about 70 kDa.
METTL3 is the catalytic subunit of the main m6A methyltransferase complex, which it forms with METTL14 and WTAP. It installs N6-methyladenosine, the most abundant internal modification of mRNA, thereby affecting mRNA splicing, export, stability and translation. METTL3 is widely studied in development, stem cell differentiation, immunity and cancer.

Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Immunogen
Synthetic peptide based on the sequence of human METTL3.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places N(6)-adenosine-methyltransferase catalytic subunit METTL3 in the nucleus, nucleus speckle and cytoplasm. Tissue expression noted by UniProt (human): Widely expressed at low level. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
