Product Overview
This rabbit polyclonal antibody detects major facilitator superfamily domain-containing protein 12 (gene MFSD12) in human samples and is validated for Western blot, IHC and ELISA. The predicted molecular weight is 52.1 kDa, and the supplier reports an observed band at about 52 kDa.
MFSD12 is a major facilitator superfamily transporter that imports cysteine into melanosomes and lysosomes. In melanocytes, this cysteine supply supports production of red-yellow pheomelanin, so MFSD12 influences skin and hair pigmentation. In other cells it helps maintain lysosomal cystine and cysteine levels, and it is studied in pigmentation and lysosome biology.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human HeLa cells, human SiHa cells, human A549 cells, human PC-3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human testicular seminoma tissue, human colon adenocarcinoma tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Recombinant human MFSD12 fragment (Gln66–His443), expressed in E. coli. Sequence identity with the mouse ortholog: 86.8%.
Reactivity Notes
The supplier lists reactivity with human. UniProt places major facilitator superfamily domain-containing protein 12 in the melanosome membrane and lysosome membrane. Tissue expression noted by UniProt (human): Widely expressed, with high expression in primary melanocytes. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
