Product Overview

This rabbit polyclonal antibody targets ubiquitin carboxyl-terminal hydrolase MINDY-3 (gene MINDY3) in human, monkey, mouse and rat samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 49.7 kDa, and the supplier reports an observed band at about 50 kDa.

MINDY-3 is a member of the MINDY family of deubiquitinating enzymes. It removes Lys48-linked ubiquitin chains from proteins, the type of chain that usually marks proteins for proteasomal degradation, and its specific cellular targets are still being defined.

Flow cytometry (fixed cells) 1–3 µg/1×106 cells (Human)
ELISA 0.1–0.5 µg/mL

Samples with a confirmed band (WB): monkey COS-7 cells, human 293T cells, human HEL cells, human K562 cells, human SH-SY5Y cells, rat kidney tissue, mouse kidney tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (ICC/IF): U2OS cells.

Samples with a confirmed signal (flow cytometry): HeLa cells.

Immunogen

Recombinant human MINDY3 fragment (Ala52–Lys396), expressed in E. coli.

Reactivity Notes

The supplier lists reactivity with human, monkey, mouse and rat. UniProt places ubiquitin carboxyl-terminal hydrolase MINDY-3 in the nucleus. Tissue expression noted by UniProt (human): Widely expressed with high levels in heart, skeletal muscle, and kidney, and low levels in liver and brain. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
